Biotin is an ingredient in many supplements that promise stronger hair, smoother skin and harder nails. Where does this popularity come from? Mainly because people with a serious biotin deficiency often have skin problems, hair loss and brittle nails. So it makes sense that supplementation could help. Is it really so?

Does biotin help the skin?

Yes - if someone has a biotin deficiency, supplementation works very well. It can then reverse skin, hair or nail problems. Sometimes the deficiency is due to genetics, long-term use of certain medications or eating large amounts of raw egg whites (which block the absorption of biotin).

Biotin - role in metabolism

Biotin plays an important role in many processes in our body - it acts as an assistant (cofactor) for enzymes that take part in energy metabolism. Here are the most important ones:

1. Glucose Production from Other Ingredients

An enzyme called pyruvate carboxylase, dependent on biotin, allows the creation of glucose from substances such as lactate, glycerol or some amino acids. This process – gluconeogenesis – is particularly important between meals or during starvation to provide energy to the brain. When biotin is lacking, there may not be enough glucose, which leads to hypoglycemia.

2. Fat Creation

Biotin helps an enzyme called acetyl-CoA carboxylase work. This creates malonyl-CoA – a basic ingredient needed to build fatty acids. This process occurs in the liver and adipose tissue, among others.

3. Breakdown of Some Proteins and Fats

Biotin also supports enzymes responsible for breaking down amino acids (e.g. isoleucine, valine, leucine) and fats with an unusual structure. This allows the body to use them as an energy source. When these enzymes do not function properly – for example, due to a biotin deficiency – harmful substances can accumulate in the body.

Dietary sources of biotin and how it is absorbed

Biotin is commonly found in many foods, both animal and plant. Good sources include liver, eggs (especially the yolk), yeast, nuts, seeds, avocados, sweet potatoes and whole grains. Some biotin can also be synthesized by bacteria in the large intestine, although the extent to which it is absorbed and utilized by the body is not fully understood.

Biotin Deficiency - Causes and Symptoms

Biotin deficiency, although rare, can be caused by a variety of factors and can manifest itself in a range of symptoms.

Causes of Deficiency

  • Congenital disorders of biotin metabolism: The most common are holocarboxylase synthetase deficiency (an enzyme that attaches biotin to carboxylases) and biotinidase deficiency (an enzyme that releases biotin from proteins and peptides). These genetically determined conditions require treatment with high doses of oral biotin.
  • Insufficient dietary intake: Extremely rare with a varied diet, but possible with long-term consumption of large amounts of raw egg albumin or in the case of total parenteral nutrition without supplementation.
  • Absorption disorders: Rarely caused by intestinal diseases, but some anticonvulsantsmay interfere with the absorption or increase the metabolism of biotin.
  • Long-term use of antibiotics: Theoretically, it may interfere with the production of biotin by intestinal bacteria, but the clinical significance of this is unclear.

Symptoms of Biotin Deficiency

Symptoms of biotin deficiency develop gradually and may include:

  • Skin symptoms: Scaly, red rashes around the eyes, nose, mouth and perineum, inflammation skin
  • Neurological symptoms: Depression, apathy, hallucinations, numbness and tingling of the limbs (paresthesia), ataxia (problems with motor coordination), seizures.10
  • Hair and nail symptoms: Hair loss, brittle nails.
  • General symptoms: Fatigue, loss of appetite, nausea, muscle pain.

Congenital deficiencies of biotin-related enzymes can lead to serious metabolic and neurological problems if left untreated.
